Heterologous expression of bacterial cytochrome P450 from Microbacterium keratanolyticum ZY and its application in dichloromethane dechlorination

Dichloromethane (DCM) is a volatile halogenated hydrocarbon with teratogenic, mutagenic and carcinogenic effects. Biodegradation generally regarded as an effective economical approach of pollutant disposal. In this study, novel strain was isolated its cytochrome P450 heterologously expressed for DCM degradation. The isolate, Microbacterium keratanolyticum ZY, characterized Gram-positive, rod-shaped flagella-existed bacterium without spores (GenBank No. SUB8814364; CCTCC M 2019953). After successive whole-genome sequencing, assembly annotation, eight identified functional genes (encoding P450, monooxygenase, dehalogenase hydrolase) were successfully cloned in Escherichia coli BL21 (DE3). recombinant expressing presented the highest degradation efficiency (90.6%). Moreover, specific activity more than 1.2 times that (from Methylobacterium rhodesianum H13) under their optimum conditions. kinetics by well fitted Haldane model value maximum rate determined to be 0.7 s−1. might occur through hydroxylation, dehydrohalogenation, dechlorination oxidation generate gem-halohydrin, formyl chloride, formaldehyde formic acid. study helps comprehensively understand process actions bacterial enzymes (cytochrome dehalogenase).
